Definition |
The traditional method widely used as a functional test for alloreactivity between patient and donor is mixed lymphocyte culture (MLC). However, this method is unable to determine either the number or the functional capacity of responding T lymphocytes and has proved insufficiently sensitive to predict the incidence and severity of GVHD after HLA-identical sibling transplantation. In mixed lymphocyte culture (MLC) lymphocytes are collected from the lymph nodes (for responder cells) and the spleen (for stimulator cells) of rats with and without transplants by centrifugation. After the lymphocytes are washed with culture medium, stimulator cells were treated with mitomycin C. The tissue culture medium (RPMI 1640, GIBCO, Grand Island, N.Y.) used in this experiment was supplemented with 2-mercaptoethanol, heat-inactivated fetal calf serum, L-glutamine and penicillin-streptomycin.
