Context Nonviral vectors are at present actively investigated as alternatives to recombinant viruses for gene transfer studies and gene therapy applications. Among them, cationic lipids are particularly attractive as it is possible to synthesize a wide variety of well-characterized reagents. Although the structure-activity relationships of cationic lipids are not fully understood, it is generally agreed that the nature of the positive headgroup impacts on their transfection activity. Thus, we have directed our efforts over the last several years toward the development of lipids with novel cationic moieties. In particular, we have recently explored the transfection potential of lipophilic derivatives of aminoglycosides.
